Introduction

AP is an extremely stable enzyme, resistant to heat denaturation and to denaturation by sodium dodecyl sulfate (SDS). It is a dimeric metalloenzyme, containing 4 Zn atoms and 2 Mg atoms per dimer. In addition it has recently been shown to possess an intense, long-lived tryptophan phosphorescence. Although AP has three tryptophans per monomer, it has been determined that the phosphorescence arises from Tryptophan 109 (W109), located near the core of the protein. It is this phosphorescence and the dependence it shows upon the environment of W109 that is our primary tool for studying the folding and stability of AP.
